Laboratory regulations



Rules for users of the NHN lab

1). No experiments can be done by new users of the NHN lab without personal guidance by an experienced NHN user the first week

2). From all DNA extracted a sample will be donated to the NHN DNA bank

3). For all DNA extracted a voucher specimen will be preserved in a public herbarium. This voucher specimen should be a fertile herbarium specimen.

4). All users not employed by the NHN are obliged to pay a benchfee (the exact amount depends on duration of stay, amount of experiments planned and financial situation of user and will be fixed by the NHN-teamleader and -management)


General rules and savety regulations

General

  • Bags and coats are not allowed in the lab; bring only the necessary things in.
  • In the lab you must wear a labcoat but especially during working with EtBr and Hazardous Chemicals. Don't place your labcoat on the lab table but put it on the rack.
  • If you wear gloves, NEVER touch taps, doorhandles, equipment, keyboards, computers, lightswitches etc with the gloved hand. If you don't trust it is "clean", then use a clean tissue to touch it.
  • It is strictly prohibited to eat, drink or smoke in the lab.
  • It is prohibited to write or stick comments on the lab equipment without permission.
  • It is prohibited to take chemicals, reagent etc from the Molecular Lab to the VMT lab or other places. If you think this is necessary, please contact the labtechnician.

Safety

  • Always read instructions on the chemicals and reagents first before using them. In case of doubt consult the labtechnician.
  • Dispose chemical/biological waste in appropraite containers (see point Waste).
  • Special attention must be given to hazardous chemicals/reagents.
  • Wash your hands before you leave the lab!

General tasks

  • Refill the MilliQ tanks when they are nearly empty.
  • Fill up pipet tips, eppendorf tubes and MilliQ bottles and autoclave. Put autoclaved material in the oven to dry. Store the materials after drying in the cupboard for autoclaved material.
  • Put clean glassware from the dry-rack in the cupboard.
  • Fill in the order list if a certain product is running out and/or inform the labtechnician if this is urgent.
  • If you are the last one leaving the lab, check if lights, radio and machines are switched off (except those which are meant to stay on).

Cleaning

  • Keep the lab tidy. Everyone has to clean his/her place/lab table before leaving.
  • Once per month there will be a labcleaning. Everyone who used the facillities of the lab that month must help to clean it.
  • Cleaning of glassware:
    Clean the glassware first 3 times with tapwater followed by three times with demi-water. Put it on the rack to dry and don't forget to put it in the cupboard when dry.
  • Cleaning of spilled EtBr:
    In case of little spilling, clean area with tissues soaked in alcohol 70%. Dispose the tissues in the special white buckets for EtBr waste. In case of large spilling, mark but avoid the contaminated area and contact the labtechnician.
  • Cleaning of spilles chemicals (especially near the balances):
    Always clean spilled chemicals immediately. In case of spilling hazardous chemicals, mark but avoid the contaminated area and contact the labtechnician.

Waste

  • Put watse in the appropraite bags/container:
    • EtBr and Acryl Amede solid waste: white bucket near Gel-Doc system/electoforese room.
    • EtBr concentrated liquid waste: black labeled tank in fume cabinet.
    • Chemical waste (liquid): labeled tanks in fume cabinet.
    • Phenol waste, i.e. epp's with phenol: phenol bucket under fume cabinet.
    • Tips without EtBr or phenol: yellow plastic buckets on table
    • Tips with EtBr: white bucket in fumecabinet and in electoforese room.
    • Buffer containing EtBr: black labeled tank in fumecabinet.

Machines

  • When you use a machine and you are not familiar with it, read the equipment instructions carefully (see map "Equipment Instructions").
  • When you want to use the ABI 377 on your own, you need permission from the labtachnician.
  • It is not allowed to install software on the computer next to the ABI 377.

Shared chemicals

  • When you use chemicals, reagent or consumables from the "shared chemical list", fill in the appropriate list in the "shared chemical database" on the lab computer.

Additional rules VMT lab

  • Every newcomer in the VMT lab must have been instructed by an authorized person about safe working and the rules concerning the VMT lab.
  • Everyone who enters the lab must record his/her time of presence and reason in the VMT logbook.
  • Everyone who is in the lab must wear a VMT labcoat (green lable) and put his/her name on the namebadge. It is prohibited to wear "own" labcoats.
  • It is prohibited to take the VMT labcoat outside the lab. Change coats regularly.
  • It is prohibited to open windows/doors to the balcony, except during an emergency.
  • It is prohibited to wear gloves during VMT work, gloves are allowed for DNA work and in case of cleaning spilled material.
  • Keep the lab tidy. Everyone has to clean his/her place/lab table before leaving.
  • In the VMT lab only work on VMT-level or lower is allowed. When working with bacteria, always clean table before and after this work with alcohol 70%. After spilling material you have to clean immediately and according to the VMT cleaning and safety rules. In case of doubt and in case of large contamination contact the labtechnician.
  • Put VMT-waste in the appropriate bags/container.
  • Used glassware and other contaminated materials must be autoclaved daily.
  • Don't work in fumecabinet during the autoclave process.
  • When you leave the lab, hang your labcoat in the locker.
  • Wash your hands before leaving the lab.
  • Correct people who don't work according these rules: it's for everyones safety!

VMT-waste

  • VMT contaminated pipettips and 1.5 ml cups: small yellow needlecontainer.
  • Biological waste, VMT waste and yellow needle containers: blue containers.
  • Other, non-VMT waste or autoclaved VMT-waste: grey bags (in big yellow container).
